General description

The new AST Assay Kit, MAK467, is now available! Aspartate Aminotransferase (AST), formerly called serum glutamate-oxaloacetate transaminase (SGOT), is a pyridoxal phosphate (PLP)-dependent enzyme that catalyzes the conversion of aspartate and α-ketoglutarate to oxaloacetate and glutamate. Similar to Alanine Aminotransferase (ALT), AST levels in blood are commonly used as a marker for liver function. However, AST has a broader tissue distribution than ALT and perturbations in AST levels can occur in response to diseases or injuries in multiple tissues including skeletal and heart.

Biochem/physiol Actions

The Aspartate Aminotransferase Activity Assay kit provides a simple and direct procedure for measuring AST activity in a variety of samples. In this kit, the transfer of an amino group from aspartate to α-ketoglutarate results in the generation of glutamate, resulting in the production of a colorimetric (450 nm) product proportional to the AST enzymatic activity present. One unit of AST is the amount of enzyme that will generate 1.0 μmole of glutamate per minute at pH 8.0 at 37 °C.

  1. Could you tell me the average number of readings or typical total incubation time (at 37 °C) until the most active sample reaches the linear range limit?

    1. The time required to complete the assay depends on the overall activity of aspartate aminotransferase in the sample. Running the assay for approximately one hour is recommended, with readings taken every 5 minutes. However, adjustments to the incubation time and reading intervals may be necessary based on the observed activity. For low activity, a longer incubation period than 60 minutes may be required, while high activity may necessitate a shorter incubation.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Is this kit only suitable for human samples? Or can I measure murine AST as well?

    1. This product has been replaced by the Aspartate Transaminase (AST) Assay Kit (MAK467). Both kits are suitable for the determination of AST activity in mammalian species other than humans.

  4. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   5. Is it possible to use the MAK055 for an experiment that involves evaluating samples with and without B6 added? Additionally, will hemoglobin interfere if a blood hemolysate sample is used when assayed with this kit?

    1. The specific experiment in question has not been performed in the lab, and there are no references for the Aspartate Aminotransferase Activity Assay Kit where a similar experiment has been reported. Considering the nature of the assay and the fact that B6 is a co-enzyme of the ASP, necessary for its activity, in principle, the kit will be suitable to evaluate samples treated with B6. It will be necessary to evaluate any possible interferences and optimize user-specific conditions.
      It is possible that hemoglobin will interfere with the assay if hemolyzed blood is tested. Blood is generally not recommended as a sample, unless specifically mentioned in the assay, due to the possible interference as mentioned. Furthermore, the kit has not been tested with blood samples.

  6. Does the AST Activity Assay Kit (MAK055), which includes a positive control (MAK055F), have a specific species origin? For instance, is it Human AST?

    1. The positive control in MAK055 is derived from porcine heart.

  7. Do you anticipate phenol red in culture medium having an impact on the colorimetric read out at 450 nm? (I'm thinking there may be some spectral overlap)

    1. Ideally, culture media should be phenol red-free.
