Gene Information

General description

The new Lactate Dehydrogenase Assay Kit, MAK464, is now available! Lactate dehydrogenase (LDH) is an oxidoreductase enzyme that catalyses the interconversion of pyruvate and lactate. Cells release LDH into the bloodstream after tissue damage or red blood cell hemolysis. Since LDH is a fairly stable enzyme, it has been widely used to evaluate the presence of damage and toxicity of tissue and cells. LDH is also elevated in certain pathological conditions such as cancer. Quantification of LDH has a broad range of applications.

Application

Lactate Dehydrogenase Activity Assay Kit has been used for measuring the activity of lactate dehydrogenase (LDH) in the cell culture medium.[1][2]
Lactate Dehydrogenase Activity Assay Kit has been used to determine the concentration of lactate dehydrogenase in samples.

Biochem/physiol Actions

In this kit, LDH reduces NAD to NADH, which is specifically detected by colorimetric (450 nm) assay. The LDH Activity Assay kit quantifies LDH activity in variety of biological samples. The assay is quick, convenient, and sensitive.

Questions

1–5 of 5 Questions
  1. How can I determine the shelf life / expiration / retest date of this product?

    1 answer
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  2. Is it possible to use cell media instead of cell (as the LDH enzyme secreted in media if the cell membrane is damaged)for determining LDH activity?

    1 answer
    1. Yes, the Lactate Dehydrogenase Activity Assay Kit is suitable for measuring the activity of lactate dehydrogenase (LDH) in cell culture medium.

      Helpful?

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 answer
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  4. Do the reagents in MAK066 Lactate Dehydrogenase Activity interact with the EDTA present in plasma samples? Alternatively, does EDTA in plasma affect the results?

    1 answer
    1. Conventional amounts of EDTA used to prepare plasma samples should not interfere with the MAK066 Lactate Dehydrogenase Activity Assay Kit in any way.

  5. Part of the kit is also the positive control. Could you provide information what are the expected readings or how it can be used as a control of the final readings or the samples.

    1 answer
    1. The LDH Positive Control included in this kit is purified lactate dehydrogenase (LDH) from rabbit. The exact concentration of the enzyme is proprietary; but each vial contains </= 1 U of enzyme. Do not compare samples to the positive control; it is not a standard. Please see the image below. It contains a graph of Kinetic profiles of approximately 0.5 mU of a sample of pure LDH (Positive Control) and 2 μL frozen human serum using buffer as a background control:
