General description

Hemoglobin is the major component of red blood cells, and is responsible for their red color. Its normal concentration in erythrocytes is 34%. Hemoglobin is the most important respiratory protein of vertebrates by virtue of its ability to transport oxygen from the lungs to body tissues, and to facilitate the return transport of carbon dioxide.

Application

Hemoglobin was used in the development of a rapid enzyme immunoassay for the detection of retinol-binding protein.[1] It was also used in the inhibition of human platelet reactivity by endothelium-derived relaxing factor.[2]

Biochem/physiol Actions

Oxygen transporter, NO scavenger
The Fe2+/Fe3+ balance is a physiological indicator of blood oxygenation. Deoxygenated hemoglobin accessorizes a feedback loop by reducing nitrite to NO, a vasodilator which enhances blood flow to oxygen-deprived tissues.

  2. Would H7379 Hemoglobin Human has population of glycated hb as well? If yes, what level?

    1. MilliporeSigma, on product H7379, does not perform any sort of characterization as to whether any particular subfraction of H7379 is glycated. H7379 is a general hemoglobin product and not any single one isoform.

  3. I would like to know if the product loses stability if it is kept in a freezer at -20, since the recommended temperature is between 2 and 8 degrees Celsius.

    1. The stability of the hemoglobin powder at -20°C has not been evaluated. However, storage of the powder at freezer temperature is not expected to cause degradation. Please see the link below to review additional information available in the product datasheet:

  4. What are the recommendations for long-term storage once it is solubilized? Is it OK to make single-use aliquots for -80°C?

    1. The stability of this material in solution has not been evaluated. However, other commercially available hemoglobin solutions, intended for use as microbiological medium supplements, are reportedly stable for more than 1 year when stored at 2-8°C.

  5. Can it be reconstituted to a final concentration of 17 g/dL?

    1. A concentration of 17 g/dL exceeds the solubility recommendation. Each batch released is tested and guaranteed to dissolve in deionized water at up to 20 mg/mL.

  6. What is this product H7379 lyophilized in? When I reconstitute in water should I account for sucrose or other buffer components?

    1. The protein was dialyzed against deionized water before the lyophilization step.

  7. I noticed that the solubility of H7379 in H2O is only 20 mg/mL. We are looking for Hb solution with concentration from 10 to 20 g/dL. Could you kindly advise whether the power can have higher solubility in any other liquid?

    1. This product is tested for solubility at 20 mg/mL during quality control testing in water. Testing in other liquids for higher solubility has not been performed.

  8. What is the standard of this reagent?

    1. This product contains a dark red to red-brown human hemoglobin powder. The standards specified in the Certificate of Analysis include low water content (≤ 15%) and an iron level within 0.25–0.35%. Tests for hepatitis B surface antigen, hepatitis C virus, and human immunodeficiency virus are negative.
