Application

Poly-L-lysine polymers can be used in promoting cell adhesion to solid substrates, conjugation to methotrexate for increased drug transport, microencapsulation of islets, cell microencapsulation technology, microarray glass slide coating, and chromosomal preparations. Lower molecular weight poly-L-lysine (30,000-70,000) is less viscuous in solution, but higher molecular weight versions provide more attachment sites per molecule.
Poly-L-lysine solution has been used for the attachment of endothelial cell cross sections to UV-crosslinked PET membrane frame slides. Poly-L-lysine coated slides were used for the placement of tissue sections for immunohistochemical assay.

Biochem/physiol Actions

Poly-L-lysine is a nonspecific attachment factor for cells useful in promoting cell adhesion to solid substrates by enhancing electrostatic interaction between negatively charged ions of the cell membrane and the culture surface. When it is absorbed to the cell culture surface, poly-L-lysine functions to increase the number of positively charged sites available for cell binding. With cells that can digest poly-L-lysine, poly-D-lysine should be used as the attachment factor.

Preparation Note

This product is supplied as a 0.10% (w/v) solution in water. Coating slides with this solution should be followed with a 5 minute incubation and drying the slides at room temperature. Coated slides will be stable for one year if protected from dust.

Other Notes

Poly-L-lysine is a positively charged amino acid polymer with approximately one HBr per lysine residue. The hydrobromide allows the poly-L-lysine to be in a crystalline form soluble in water. A small amount of product may be found in the beta structure because the HBr interferes with hydrogen bonding between amino and either the carboxyl groups or N or O containing moieties.

  2. Can I add this to maxisorp plates surfaces to make DNA binding stronger.

    1. The Poly-L-Lysine solution (P8920) was developed and tested to improve tissue adhesion to glass slides for histological and immunohistochemical use. Although other uses of P8920 have been reported, there is no data on hand regarding the use of P8920 to make the DNA binding stronger on polystyrene MaxiSorp plates.

    1. Solutions of this product are not sterile and contain a preservative, ProClin, to control contamination. In theory you could sterile filter P8920 through low protein binding filters, such as PVDF or PES. Sterile filtering will not remove the ProClin from P8920. Live cells should not be exposed to ProClin, as ProClin interrupts the Krebs cycle at 4 different sites. It would be best to purchase the powder as P1524 and then sterile filter after the solution has been prepared.

    1. The number of slides that may be coated is dependent upon the surface area. The general use instructions recommend using 0.5 - 1.0 mL per 25 cm2 coating area - the 25 cm2 value is based on a T-25 tissue culture flask. A standard microscope slide has an approximate area of 1.3 cm2.

  5. What are the recommended uses for Product P8920, Poly-L-lysine solution?

    1. P8920 was developed for use in coating slides intended for use in either histology or immunohistochemistry.  Poly-L-lysine is preferred for uses such as silver staining procedures that require an alkaline pH.  Silane coated slides may be more effective is a low pH is required.

  7. Is Product P8920, Poly-L-lysine solution, recommended for cell culture use?

    1. No, the product contain 0.01% thimerosal as a preservative. For cell culture use, product P4832 is recommended.  This solution has the same molecular weight as P8920 but contains no preservative.  P4832 is sterile filtered and is considered suitable for cell culture use.
