Questions

  1. I bought Carbol-Fuchsin solution according to Ziehl-Neelsen (SKU 21820-1L) from Merck for detection of mycobacterium on tissue sections. No bacteria detected, even in the positive control slides. Is this the right reagent? Can you recommend me a protocol?

    1 answer
    1. Carbol-Fuchsin solution according to Ziehl-Neelsen, catalog number 21820, should stain acid fast mycobacterium. Quality Control testing performed for product 21820 does not include staining tissue sections. The procedure recommended by the manufacturing site is for smears. The recommend procedure can be found in Staining Procedures: Fourth Edition by George Clark, Williams & Williams, Baltimore 1981 page 380-381.

Products are guaranteed 1 year from the date of shipping. If no staining is observed, please contact your local Technical Support team to file a formal complaint.

There are other Carbol Fuchsin solutions that are performance tested during quality testing to ensure acid fast bacillus organisms are demonstrated on tissue sections. Tissue Control slides are also sold as catalog number TTR001-25ea. Catalog numbers for the Carbol Fuchsin are HT8018-250ml or HT80116-500ml. The product numbers for the counterstain for the carbol fuchsin solution are HT8028-250ml or HT80216-500ml. Instructions For Use are found on the website for the Carbol Fuchsin solutions and the Malachite Green counterstain.
