General description
Amicon® Ultra 4 mL centrifugal devices with Ultracel® 100,000 Dalton molecular weight cutoff filters provide fast ultrafiltration, and reliable retention of macromolecules. For example, IgG (156 kDa) at 1 mg/ml initial concentration is retained at >95% .
The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.
The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon® Ultra 15 mL filters deliver the highest recovery with short spin times.

What is the maximum recommended starting protein concentration from Vero cell lysate to avoid overwhelming the filter?

There is no maximum protein load for these devices, and optimal protein concentrations must be determined empirically. We do recommend a clarifying spin prior to filtration to bring down larger particulates.

Can I use Amicon Ultra 100 kDa filter only for protein sample filtration, to avoid concentration or sample purification?

This device is intended for use as a protein concentrator. Proteins above 100 kDa will be retained while smaller proteins will pass through to the filtrate fraction. Other particles within the sample may be present in both the filtrate and the retentate, depending on the nature of the particle, the nature of the filter membrane, and the centrifugation protocol or technique used.
