General description

Chromocult® Coliform Agar is a granulated, less selective, and differential chromogenic culture medium intended for microbiological analysis of food and water. This medium saves time by enabling the simultaneous detection, differentiation, and enumeration of E. coli and coliforms from drinking water and processed food matrices within 24 hours. This agar is the only chromogenic culture medium which was used for validation to prepare the new ISO 9308-1 (2014) version. The medium contains Tergitol 7 as an inhibitor of Gram-positive bacteria without any impact on the growth of the targeted coliforms/ E. coli. Chromocult® Coliform Agar is, therefore, the ideal medium for the detection of coliforms/ E. coli in drinking water and processed foods. It facilitates and speeds up your processes.
E.coli / Coliform Selective-Supplement (1.00898) is available to enhance selectivity and growth conditions.

Application

Chromocult® Coliform Agar is used for the simultaneous detection of coliform bacteria and E. coli in drinking water, disinfected pool water, finished water from treatment plants and processed food samples.

    1. Preparation instructions are as follow:

      Dissolve 26.5 g in 1 liter of purified water. Heat in boiling water and agitate frequently until completely dissolved. Do not autoclave, do not overheat!

      Cool the medium to 45 - 50 °C and pour plates. According to EN ISO 9308-1 dispense in Petri dishes to a depth of at least 4 mm (approximately 18 ml in a 90 mm plate).

      For using by poured plate method cool the medium to 44 - 47 °C in a water bath before use (see EN ISO 11133).

      The prepared medium is slightly turbid to turbid and yellowish. The pH value at 25 °C is in the range of 6.6-7.0

      There should be no visible moisture on the plates before use. When moisture is present, the plates should be dried for the minimum time required to remove visible moisture, following the procedure as described by EN ISO 11133.

      Preparation and addition of optional Cefsulodin solution (US-EPA approved method): Dissolve 5 mg of Cefsulodin in 2 ml of purified water and sterilize by membrane filtration (0.2 μm nominal pore size). Aseptically add the solution (2 ml per 500 ml of medium) to 500 ml of liquefied medium (45 - 50 °C). Pour plates as described above.

  2. What precautions do you need to take to produce chromocult Coliformes agar, since it cannot be overheated or autoclaved? What are the recommendations? Sterile water? Sterile glassware?

    1. Ideally, a boiling hot water bath would be best. However, you may also use a hot plate with a magnetic stirrer to prepare the medium. Using a magnetic stir bar, thoroughly mix the powdered media with purified water until no clumps are visible. Use a container larger than the volume being prepared (for example - 1L Erlinmeyer flask to prepare 500 mL media). Heat media to 100°C, which will be the boiling temperature. Boiling will melt the agar, but bubbling will occur. Closely monitor to prevent boiling over. Media is sufficiently heated when the appearance transitions from cloudy/hazy to transparent. Allow media to cool to 45 - 50°C and pour aseptically onto plates. For larger volumes, the media can be dispensed by using a peristaltic pump, Sterile plates, utensils, and/or tubing may be used during the preparation and filling process.
