General description
Plaqueamento de células alimentadoras MEF
Procedimento
1. Antes de descongelar as células alimentadoras PMEF, cubra as placas/frascos com solução de gelatina.
2. Descongele o(s) frasco(s) de PMEF rapidamente em banho-maria a 37 °C e transfira para um tubo de 15 ml (já contendo 10 ml de meio de célula alimentadora de PMEF quente). Inverta suavemente o tubo para distribuir e centrifugue a 300 xg durante 4–5 minutos.
3. Remove o sobrenadante e ressuspende o sedimento celular em meio de célula alimentadora de PMEF quente.
4. Remove a solução de gelatina das placas/frascos e separe alíquotas da suspensão de células alimentadoras de PMEF nas densidades recomendadas na Tabela 4.1 do guia de protocolo ES do camundongo.
5. Incuba as células alimentadoras de PMEF a 37 °C com 5% de CO2. Use as Figuras 4A, B e C no guia de protocolo ES do camundongo como um guia para uma estimativa da densidade e
aparência correta do PMEF. Placas gelatinizadas podem ser utilizadas por 12–14 dias.
