Depending on the staining target, the preparation of the dye solution can vary. One method involves dissolving the dye at 0.01% in water. Other method uses a 2% concentration in water. For mast cells, another procedure involves using 0.02 grams of the dye, dissolving it in 100 ml of water, and adding 0.25ml of glacial acetic acid. In electron microscopy, two solutions are made: Solution 1 (2 grams of dye dissolved in 100 ml of water) and Solution 2 (2 grams of sodium borate, also dissolved in water). Equal parts of Solution 1 and Solution 2 are used for staining. Not all lots of dye are soluble to 2% in water. T3260 is tested for solubility at 4mg of dye in 4 ml of water, or 1 mg/ml. Point to note is that whether it will provide adequate staining needs to be determined since it hasnot been involved in staining actual cells or tissue sections.

Toluidine Blue O and Toluidine Blue are generally regarded as synonyms. The dye can be found in 2 forms. There is a chloride form and zinc chloride double salt. Products T3260 and 198161 are the chloride form. This is the form certified by the Biological Stain Commission. The dye listed as simply Toluidine Blue, Product 89640, is the zinc chloride double salt. For routine use today, the chloride form should be utilized. The product number recommended for biological/histological staining is Product 198161, Toluidine Blue O. This product has been found to provide acceptable staining of tissue by the Biological Stain Commission. Products 198161 and 89640 have not been suitability tested for the staining of tissue sections.
