Histological analysis of bone encased tissue is severely hampered by technical difficulties associated with sectioning calcified tissue. Cryosectioning of bone is possible but requires significant technical adaptation and expensive materials and is often time-consuming. Some decalcifying reagents in common use

Questions

  1. I’m using a decalcification solution, and the solution suddenly turned black, almost like diluted paint. I don’t remember it being this dark before. Is the precipitate normally black, or does this indicate some kind of problem?

    1 answer
    1. Decalcifying Solution-Lite will discolor and corrode most metals. Avoid exposure to metal cassettes, countertops and slide racks. When rinsing specimens, flush any Decalcifying Solution-Lite with running water to prevent damage to chrome plated plumbing fixtures. All decalcifying must be performed in plastic (or glass) containers.
