General description

Pellet Paint NF Co-Precipitant is a nonfluorescent dye-labeled carrier compatible with fluorescent sequencing. It facilitates rapid removal of BigDye Terminators during the alcohol precipitation of cycle sequencingreaction products. Cycle sequencing reactions can be precipitated rapidly with 1 µl of carrierper reaction and centrifugation times of 10minutes. The easily visualized carrier providesa simple confirmation that precipitation has occurred. Sequencing reaction products are efficiently pelleted and dye-labeled terminators remain in the supernatant during alcohol precipitation using the standard Applied Biosystems precipitation protocols. Resuspension of pelleted sequencing reaction products in deionized formamide can beconfirmed by checking for dissolution of the carrier. Pellet Paint NF Co-Precipitantis fully compatible with the ABI PRISM®BigDye Terminator Cycle Sequencing Ready Reaction. To avoid extra sample handling, Pellet Paint NF Co-Precipitant can be added directly to the reaction mix, template DNA, crude PCR samples, or dilution buffer before the cycle sequencing reaction. Although PelletPaint NF absorbs in the UV range, accurate spectrophotometric measurements of DNA or RNA samples are possible; the absorbance ratio (provided with each package of Pellet PaintNF) can be used as a correction factor when determining nucleic acid concentration. PelletPaint NF Co-Precipitant has no detectable effect on the sequencing reaction or sequence accuracy. Pellet Paint NF Co-Precipitant is a useful substitute for the original Pellet Paint Co-Precipitant in applications where fluorescent detection is used.

  1. What is the recommended retest method to verify product quality after passing the recommended retest date?

    1. On products, we occasionally publish a retest date.  This is a date, created years after manufacturing dates, which is meant for internal use only.  This date is chosen if we still have inventory of a specific lot years later.  If we have inventory of that lot, we decide to discard remaining product or retest it internally if there is a good amount of product remaining that may still be stable and of good quantity years later.  Retest instructions and protocols are only meant for internal use.  If a product is retested years later, and passes, and continues to stay in inventory, we issue a new Certificate of Analysis/Quality with a second, new date, years in the future.

  2. Is there a lower limitation of DNA fragment size for use with the Pellet Paint® NF Co-Precipitant 70748? I have DNA fragments at around 500 bp.

    1. there is no recommended lower limitation of DNA fragment size for the Pellet Paint NF Co-Precipitant 70748. However, it notes that precipitation based on ethanol method may lead to lower yield with small DNA fragments, but using DNA fragments at 500 bp should pose no problem.
