Sigmaのα-キモトリプシンは、昆虫Locusta migratoriaに由来する2つの同種の阻害剤（pars脳間部主要ペプチド-Cおよびpars脳間部主要ペプチド-D2v）の、酵素との複合体形成による結晶構造の解析に使用されています。[1]
Biochem/physiol Actions
Α-キモトリプシンは241個のアミノ酸からなるセリンペプチダーゼであり、ジスルフィド結合によって連結した3つのポリペプチド鎖（A鎖-13残基、B鎖-131残基、およびC鎖-97残基）からなります。本酵素の分子量は、25 kDaであることが分かっています。pIは8.75です。本酵素は、チロシン、フェニルアラニン、トリプトファン、およびロイシンのC末端側のペプチド結合を選択的に加水分解します。Ca2+は、酵素を活性化および安定化します。本酵素は、ジイソプロピルフルオロホスフェート（DFP）、フェニルメタンスルホニルフルオリド（PMSF）、N-p-トシル-L-フェニルアラニンクロロメチルケトン（TPCK）、キモスタチン、アプロチニン、α1-アンチトリプシン、α2-マクログロブリン、10 mM Cu2+およびHg2+によって阻害されます。
結合のカルボキシル基側に芳香族側鎖または大きな疎水性側鎖(Tyr、Trp、Phe、Met、Leu)を持つペプチド結合を加水分解するセリンプロテアーゼです。

TLCK処理は、キモトリプシン中に存在している可能性のあるトリプシンを不活化し、キモトリプシンの活性には影響しません。
