General description

Flotillin 2 (C-terminal) encodes a caveolae-associated, integral membrane protein, which is thought to function in neuronal signaling.
Flotillin-2 is a protein encoded by the FLOT2 gene in humans. Flotillin 2 is more widely distributed, but is absent in skeletal muscle.[1] FLOT2, also called reggie-1, belongs to flotillin protein family integral membrane proteins comprising of prohibitin homology (PHB) domain or SPFH domain (stomatin/prohibitin/flotillin/HflK/C domain).

Immunogen

synthetic peptide corresponding to amino acid residues 415-428 of human flotillin 2 with an N-terminal added cysteine, conjugated to KLH. The corresponding sequence differs by 2 amino acids in rat and mouse.

Application

Anti-Flotillin 2 (C-terminal) antibody produced in rabbit has been used in:
  • immunofluorescence
  • immunoprecipitation
  • immunoblotting[2]

Biochem/physiol Actions

Flotillin-2 is a protein encoded by the FLOT2 gene in humans. Flotillins are involved in cell adhesion processes and play a role during breast cancer progression. FLOT2 especially plays a functional role in cell-cell adhesion in non-malignant epithelial cells.[3] It may have an essential function in intestinal physiology, especially in the colon. FLOT2 has been implicated in several signaling pathways in tumor cells and can be used as a prognostic biomarker for breast cancer progression.[4]
The prohibitin homology (PHB) domain constitutes a lipid recognition motif which can target proteins to the plasma membrane. The association of flotillin 2 to the plasma membrane depends on palmitoylation and myristolation in a conserved Cys34. Flotillin 2 overexpression is associated with melanoma progression and transformation of SB2 melanoma cells to a highly metastatic line. It is also associated with expression of the thrombin receptor, protease activated receptor 1 (PAR-1), that is implicated in cell growth and metastasis. Flotillin 2 may be used as a marker protein for lipid raft microdomains.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
