Application

Calcein has been used:
  • in whole cell staining
  • in histomorphometry and micro-computed tomography
  • to label active bone-forming surfaces

Biochem/physiol Actions

Calcein is a calcium-dependent fluorescent molecule. It is mainly required to study bone metabolism under in vivo conditions and helps in the staining of pit area under in vitro conditions. Calcein is used for the fluorometric determination of calcium and EDTA (ethylenediaminetetraacetic acid) titration of calcium.

Domande

1–6 di 6 domande
  1. Is this calcein (C0875) membrane-permeable or membrane-impermeable?

    1 risposta
    1. No in-house testing has been performed to determine if Calcein is membrane permeable or membrane impermeable. The literature suggests Calcein appears to be membrane impermeable. Conn's Biological Stains, 10th Edition, page 250 states, "entry of the dye into cells may be achieved by direct insertion of the hydrophilic calcein, or by the uptake of calcein AM with intracellular formation of calcein."

  2. What is the Department of Transportation shipping information for this product?

    1 risposta
    1. Transportation information can be found in Section 14 of the product's (M)SDS.To access the shipping information for this material, use the link on the product detail page for the product.

      Utile?

  3. What are the excitation and emission wavelengths of Calcein, Product C0875?

    1 risposta
    1. Conn's Biological Stains, 10th Edition states excitation is at 494 nm in aqueous solutions at pH 9; the emission maximum is 517 nm. Depending upon the pH of the solution, slightly different values have been reported in the literature.

      Utile?

  4. What is the solubility of Calcein, Product C0875?

    1 risposta
    1. Calcein is soluble at a concentration of 50 mg/mL in 1M sodium hydroxide, yielding clear orange to brown solution.

      Utile?

  5. Is there a reference for using Calcein, Product C0875, to study bone growth?

    1 risposta
    1. Clement-Lacroix, P., et al. Osteoblasts are a new target for prolactin: analysis of bone formation in prolactin receptor knockout mice. Endocrinology, 140, 96-105 (1999).

      Utile?

  6. What is the extinction coefficient of Calcein, Product C0875?

    1 risposta
    1. For samples in 1 M sodium hydroxide: E1% = 1040 at 500-504 nm; E1% = 466 at 242-245 nm.
