Soybean trypsin inhibitor is a mixture of the three types. We have not characterized our trypsin inhibitor products in terms of how much of each of the three types may be present. All three have a molecular weight of between 20,000 and 20,200 daltons, so one might use 20,100 daltons as an average.The three types vary in the amino acids at a few positions. The detailed summary of the three types (or variants) was published in the following linked reference: J. Biochem. (Japan), 98, 435-448 (1985).Note that the authors use Tia, Tib and Tic to indicate the three variants.

Questo inibitore agisce nei confronti della tripsina e, in misura minore, della chimotripsina e della plasmina. Inibisce anche le proteasi con meccanismi simili alla tripsina, la callicreina plasmatica e il fattore X della coagulazione. L′inibitore della tripsina non è attivo nei confronti delle metalloproteasi, della callicreina tissutale, delle proteasi acide o delle tioproteasi. Questo inibitore agisce formando un complesso stechiometrico 1:1 con il sito attivo della proteasi e quindi scindendo un singolo legame arginina-isoleucina sull′inibitore. L′inibizione è reversibile e dipendente dal pH.
Preparation Note
L′inibitore della tripsina è solubile in acqua e tamponi fosfato 10 mg/mL. È solubile in soluzioni saline bilanciate a 1 mg/mL e in terreni privi di siero. Soluzioni concentrate oltre 10 mg/mL possono essere leggermente torbide e di colore da giallo ad ambrato. Dopo la tripsinizzazione delle cellule, risospendere in 1 mL di soluzione di inibitore della tripsina a 1 mg/mL per ogni mL di soluzione di tripsina usata per la dissociazione. La sospensione cellulare deve quindi essere centrifugata a 1.000 rpm per formare un pellet cellulare.
Le soluzioni possono mantenersi attive se conservate a 2-8 °C a breve termine. Le soluzioni sono stabili in aliquote congelate a -20 °C.
