General description

Wiskott-Aldrich Syndrome Protein and SCAR Homolog (WASH) is a novel protein belonging to the WASP family, a group of nucleation promoting factors (NFPs) that activate a critical component in endocytosis, Arp 2/3. WASH contains three specific domains: a C-terminal VCA domain, a proline-rich domain, and an N terminal region containing two WASH homolog regions, WHD1 and WHD2. Although WASH has yet to be completely characterized, it is known to function as an NPF at the endosomal surface, where it is involved in the recruitment and subsequent activation of Arp2/3 in order to initiate actin polymerization. Therefore, WASH has a crucial role in endosomal sorting as a mediator of tubule fission resulting from Arp2/3 activation.
