General description

Il DMEM (terreno di Eagle modificato da Dulbecco), un terreno per colture cellulari usato che si usa regolarmente, è una fonte ricca di energia che contiene glucosio, vitamine e amminoacidi.[1] La miscela 1:1 di terreno DMEM e miscela di nutrienti Ham′s F-12 è molto usata nella ricerca sulle colture cellulari. A compensazione della perdita di capacità tampone causata dall'assenza di siero, alla formulazione si può aggiungere il tampone HEPES (4-(2-idrossietil)-1-piperazineetansolfonico) alla concentrazione finale di 15 mM.

Application

Il terreno DMEM con miscela di nutrenti Ham′s F-12 è stato usato:
  • come componente nel terreno di coltura di cellule stellate epatiche umane attivate da colture primarie[1]
  • per la coltura di biopsie di cartilagine[2]
  • per la propagazione di Cowdria ruminantium[3]

  2. Is the glutamine that has already been added stable glutamine? How long is the shelf life?

    1. This product does not contain Alanyl-glutamine, which is considered a more stable form It has been supplemented with L-glutamine and will remain stable through out the shelf life of the product with proper storage and handling. Typical lots of D8437 are assigned an expiration date of 1 year from the date of release. Note that under culture conditions of 37 deg C the L-glutamine will gradually degrade.     1. This is a 1:1 mixture of the DME and F-12. The optimal carbon dioxide required for DME is 10% and for F-12 is 5%. Since this medium is a mixture, the optimal carbon dioxide concentration is typically 5% to 8%.

    1. When DME/F12 is used under serum free conditions, supplementation is necessary. DME/F12 can be used for growing the epithelial cell line TM4 (mouse testes) when supplemented with insulin, transferrin, EGF, FSH, somatomedin C, ovine growth hormone, and retinoic acid. Primary cultures of Sertoli cells can also be grown in the above supplemented media. Rat glial C6 cells have been grown in DME/F12 supplemented with transferrin, insulin, LH, and rat submaxillary gland extract. See: Jakoby, W.B., and Pastan, I.H., Cell Culture, Methods in Enzymology, 58, 103-106 (1979). (This book is listed as Product No. C0541).
