General description
OptiPrep™, o iodixanolo, è un mezzo a gradiente di intensità iodato, non ionico, caratterizzato da bassa osmolarità. Consiste in una soluzione sterile, testata per l'assenza di endotossine, contenente iodixanolo 60% (p/v) e destinata all'isolamento in vitro di particelle biologiche. Trova uso per il frazionamento e la separazione di cellule e organuli subcellulari.[1]
Application
OptiPrep™ è stato usato per isolare molecole, cellule e organuli subcellulari, quali DNA, citochine, lisosomi, reticolo endoplasmatico, perossisomi, linfociti T e cellule natural killer.[1] Inoltre, è stato usato nelle seguenti applicazioni:
- analisi dell'efficienza dell'incapsulamento delle singole cellule;[1]
- isolamento secondo gradiente di cellule neoplastiche senescenti;
- isolamento di virioni non incapsulati ottenuti da lavaggio broncoalveolare;
- isolamento di lipoproteine e macromolecole;
- purificazione di virioni
- purificazione di cellule neuronali

Why choose Product D1556, OptiPrep, instead of other density gradients such as Percoll or Histopaque?

Percoll and Histopaque must be removed prior to use through washing. If Percoll and Histopaque are not removed, these density gradients can be toxic to mammalian cells. In comparison, OptiPrep has no potential for bioaccumulation and can be considered nontoxic to cells. OptiPrep also has lower viscosity as compared to polysucrose based products such as Histopaque or Ficoll.
