Application
Amyloglucosidase from Aspergillus niger is used to hydrolyze α-D-glucosides. It may be used in the brewing of beer and in the production of bread and juices. Amyloglucosidase, from Sigma, has been used to hydrolyze glycogen into glucose monomers in order to study lipid accumulation in skeletal muscle [1].
Biochem/physiol Actions
Amyloglucosidase from Aspergillus niger is capable of hydrolyzing the α-D-(1-4), the α-D-(1-6), and the α-D-(1-3) glucosidic bonds of oligosaccharides.[2] Amyloglucosidase is an extracellular enzyme that converts starch to dextrins and glucose. The enzyme is used in the starch-processing industry in the commercial production of D-glucose from corn syrup.[3]

One unit will liberate 1.0 mg of glucose from starch in 3 min at pH 4.5 at 55 °C.

The stability of this product in solution has not been evaluated. However, 172426 Glucoamylase (syn. Amyloglucosides) from Rhizopus, notes that solutions are stable when stored frozen in aliquots at -20°C for up to 3 months. Similar stability would be expected for A7420. Avoid freeze/thaw cycles of solutions.

The starting material for Product No. A7420 is Product No. A3042, which is a preparation of the enzyme in a 1 M glucose solution. This is dialyzed against water until the glucose concentration is less than 0.02%, and then lyophilized.
