Il kit è corredato dai protocolli validati per l′estrazione e l′amplificazione del DNA genomico da code di topo, capelli, tessuti animali, saliva e tamponi buccali. Nella procedura tipo, il DNA genomico viene estratto da un campione che è stato incubato nella soluzione di preparazione dei tessuti e nella soluzione di estrazione per 10 minuti a temperatura ambiente. Il campione viene riscaldato a 95 °C per tre minuti e successivamente miscelato con una terza soluzione per neutralizzare le sostanze inibitrici prima della PCR. Una parte dell′estratto di DNA viene quindi aggiunta a una reazione PCR contenente i primer e la miscela per PCR ReadyMix REDExtract-N-Amp, inclusa nel kit.

Is the extract compatible with SYBR green qPCR?

The extract from using the XNAT kit can be used for SYBR green qPCR, but we recommend using product S4320, Extract-N-Amp SYBR Green PCR ReadyMix (https://www.sigmaaldrich.com/product/sigma/s4320_. This is a 2X reaction mix containing SYBR Green, buffer, salts, dNTPs, Taq polymerase and JumpStart™ Taq antibody. It is optimized specifically for use with the extraction reagents and contains JumpStart Taq antibody for hot start PCR to enhance specificity and SYBR Green I to act as a nonspecific reporter for real-time PCR. How do I quantitate my DNA with XNAT, REDExtract-N-Amp® Tissue PCR Kit?

The DNA extract generated using the Extract-N-Amp kit is very crude, and it is not possible to obtain a reliable 260/280 ratio. For quantitation of pure DNA, we recommend using one of our GenElute genomic DNA kits.

What is the shelf life of the kit XNAT, REDExtract-N-Amp® Tissue PCR Kit and how should I store it?

The shelf life of XNAT has not been determined. However, based on our storage guidelines, it should be usable for 1 year from the date of receipt. The REDExtract-N-Amp Tissue PCR Kit can be stored at 2-8°C for up to three weeks. For long-term storage, greater than three weeks, -20°C is recommended. Do not store in a "frost-free" freezer. We recommend to store in 10 reaction aliquots.

I am unable to detect any PCR product with Product XNAT, REDExtract-N-Amp® Tissue PCR Kit.

From XNAT Technical Bulletin, troubleshooting guide table: PCR reaction may be inhibited due to contaminants in the tissue extract. Dilute the tissue extract with a 50:50 mix of extraction and neutralization solutions. To test for inhibition, include a DNA control and/or spike a known amount of template (100-500 copies) into the PCR along with the tissue extract.
