Ogni mL di adiuvante di Freund completo contiene 1 mg di Mycobacterium tuberculosis (H37Ra, ATCC 25177), termofissato ed essiccato, 0,85 mL di olio di paraffina e 0,15 mL di mannide monooleato.
Application
Gli adiuvanti di Freund possono essere utilizzati per produrre emulsioni acqua in olio di immunogeni. Gli antigeni contenuti nelle emulsioni acqua in olio stimolano risposte anticorpali elevate e di lunga durata, imputabili al lento rilascio dell′antigene.
Gli antigeni (preferibilmente in soluzione salina) vengono generalmente miscelati con un volume uguale di adiuvante per formare un′emulsione.[1]
Biochem/physiol Actions
I micobatteri presenti nell′adiuvante di Freund completo attraggono i macrofagi e altre cellule nella sede di iniezione, rafforzando così la risposta immunitaria. Per questa ragione, l′adiuvante di Freund completo viene utilizzato per le iniezioni iniziali, mentre l′adiuvante di Freund incompleto è destinato ai richiami successivi.
