General description
Piastratura dello strato di MEF
Procedura:
1. Prima di scongelare i PMEF, ricoprire le piastre/fiasche con una soluzione di gelatina.
2. Scongelare rapidamente le vial di PMEF in un bagno ad acqua a 37 °C. Trasferirle in una provetta da 15 mL pre-riempita con 10 mL di PMEF Feeder Cell Medium pre-riscaldato. Capovolgere delicatamente la provetta per mescolare e centrifugare a 300 xg per 4–5 minuti.
3. Rimuovere il surnatante e risospendere il pellet in PMEF Feeder Cell Medium pre-riscaldato.
4. Rimuovere la soluzione di gelatina dalle piastre/fiasche e aliquotare la sospensione di PMEF alle densità consigliate nella tabella 4.1 della guida ai protocolli per ES di topo
5. Incubare le cellule nutrici PMEF a 37 °C con il 5% di CO2. Utilizzare le figure 4A, B e C riportate nella guida ai protocolli per ES di topo come riferimento per valutare la densità e l′aspetto ottimali
Le piastre ricoperte con la gelatina possono essere utilizzate per 12–14 giorni.
