General description
Lactoperoxidase ist ein wichtiges Enzym, das in Rindermilch vorkommt und zur Peroxidasefamilie gehört. Es kommt sowohl in Pflanzen als auch in Tieren vor. Die Enzymstruktur umfasst eine einzelne Polypeptidkette, die aus 612 Aminosäureresten besteht.[1]
Application
Lactoperoxidase aus Rindermilch wurde als Standard für Milch-Lactoperoxidase verwendet, um die Rolle von H2O2 bei der Hemmung des Wachstums von Staphylococcus aureus durch Lactococcus garvieae in Gegenwart von Lactoperoxidase in Rohmilch zu untersuchen.[2] Sie wird auch für die Lactoperoxidase-vermittelte Iodierung eingesetzt, um radioaktiv markierte Peptide herzustellen.[3]
Biochem/physiol Actions
Lactoperoxidase katalysiert die Oxidation von Iodid zu Iod durch Wasserstoffperoxid. Diese Aktivität bietet sich als milde, spezifische Alternative zu Chloramin T bei der Radiojodmarkierung von Proteinen und DNA an.
Lactoperoxidase trägt zum antimikrobiellen System von Milch bei,[2] indem sie ein breites Spektrum von Mikroorganismen inaktiviert. Dieses Lactoperoxidase-vermittelte antimikrobielle System wurde auch in menschlichen Sekreten wie Tränenflüssigkeit, Speichel und Milch identifiziert. Lactoperoxidase katalysiert die Oxidation von Molekülen, indem H2O2 freigesetzt wird. Das Produkt weist eine antimikrobielle Aktivität auf.[1]
