General description

Lectin from Bandeiraea simplicifolia (Griffonia simplicifolia) has A and B subunits, which recognizes αGalNAc and αGal-end groups respectively. Lectins are extracted from the seeds of the legume shrub Griffonia simplicifolia, which are carbohydrate binding proteins.

Application

Lectin from Bandeiraea simplicifolia (Griffonia simplicifolia) has been used for immunohistochemistry[1] and immunofluorescence staining.

BS-I hat eine hohe Affinität für terminale α-D-Galactosylreste mit einer sekundären Affinität für terminale N-Acetyl-α-D-Galactosaminylreste.

Lectin from Bandeiraea simplicifolia (Griffonia simplicifolia) agglutinates human blood group A and B cells. It is specific for αGalNAc- and αGal. It acts as a reagent for the detection of this αGal epitope in biological materials. Lectin from Bandeiraea simplicifolia is used as a marker for xenoreactive antigen (αGal1−3Gal) and cancer. Lectin plays an important role in fertilization, embryogenesis, inflammation, metastasis and host-parasite recognition.

Analysis Note

Die Agglutinationsaktivität wird in μg/ml ausgedrückt und aus seriellen Verdünnungen einer 1-mg/ml-Lösung, die phosphatgepufferte Kochsalzlösung mit pH-Wert 6.8 enthält, für jedes Lektin, Calcium, Magnesium und Mangan in verschiedenen Konzentrationen bestimmt. Diese Aktivität ist die niedrigste Konzentration, bei der eine 2%-ige Suspension entsprechender Erythrozyten nach einstündiger Inkubation bei 25 °C agglutiniert.

BS-I ist ein tetrameres Lektin und besteht aus den zwei Untereinheiten A und B. Es gibt fünf BS-I-Isolektine mit verschiedenen Untereinheit-Zusammensetzungen: BSI-B₄, BSI-AB₃, BSI-A₂B₂, BSI-A₃B und BSI-A₄. BSI- B₄ ist Blutgruppen-B-spezifisch und hat eine exklusive Affinität für terminale α-D-Galactosylreste, wogegen BSI-A₄ Blutgruppen-A-spezifisch ist und eine hohe Affinität für terminale N-Acetyl-α-D-Galactosaminylreste aufweist.
