Beschreibung

Immunogen

Ein 12 AS großes Peptid am C-Terminus des UCP1 der Maus/Ratte (Bouillaud et al. 1986; Ridley et al. 1986; Kozak et al. 1988; Cassard et al. 1990; Miroux et al. 1993).

Application

Dieser Anti-Uncoupling Protein 1-Antikörper ist zur Verwendung in ELISA und WB für den Nachweis von Uncoupling Protein 1 validiert.

Western-Blot: 1–10 μg/ml unter Anwendung der Chemilumineszenz-Methode. Ein gegen dieses Epitop hergestellter Antikörper hat ein Protein von ~32 kDa in braunem Fettgewebe nachgewiesen (Feil und Rafael 1994).

ELISA: 1:10.000–1:100.000 unter Verwendung von 50–100 ng UCP1/Well.

Optimale Arbeitsverdünnungen müssen vom Endanwender bestimmt werden.

Biochem/physiol Actions

Uncoupling Protein 1 (UCP1), Maus. UCP1 der Maus/Ratte ist ein mitochondriales Entkopplungsprotein mit 307 Aminosäuren (Bouillaud et al. 1986; Ridley et al. 1986; Kozak et al. 1988; Cassard et al. 1990; Miroux et al. 1993). Es ist nur zu 59 % homolog mit UCP2 (Fleury et al. 1997). Humanes UCP1 befindet sich auf Chromosom 4 (Bouillaud et al. 1986; Ridley et al. 1986; Kozak et al. 1988; Cassard et al. 1990; Miroux et al. 1993). UCP1 wird primär im braunen Fettgewebe exprimiert.

Der Antikörper wurde noch nicht auf Kreuzreaktivität mit anderen Spezies getestet. Die für die Immunisierung verwendete Sequenz weist eine 100%ige Homologie mit UCP1 der Ratte, 91 % mit UCP1 vom Goldhamster, 83 % mit UCP1 vom Kaninchen und 75 % mit UCP1 vom Menschen und Hund auf.

Physical form

Affinitätsgereinigtes Immunglobulin. Flüssig in PBS mit weniger als 0,1 % Natriumazid.
