Pepsinspaltung kann zur Herstellung von F(ab′)2-Antikörperfragmenten eingesetzt werden. Pepsin unter: www.sigma-aldrich.com/enzymeexplorer.

Biochem/physiol Actions

Bevorzugte Spaltung: hydrophobe und aromatische Reste in den Positionen P1 und P1′. Spaltet Phe-Val-, Gln-His-, Glu-Ala-, Ala-Leu-, Leu-Tyr-, Tyr-Leu-, Gly-Phe-, Phe-Phe- und Phe-Tyr-Bindungen in der β-Insulinkette auf.

Analysis Note

Der optimale pH-Wert beträgt 2–4. Aktiv in 4 M Harnstoff und 3 M Guanidin HCl. Stabil bei 60 °C. Pepsin wird bei pH-Werten zwischen 8,0 und 8,5 irreversibel inaktiviert.

Other Notes

1 U entspricht der Menge an Enzym, welche die Absorption bei 280 nm bei einem pH-Wert von 2,0 und 37 °C je Minute um 0,001 erhöht (Hämoglobin, Kat.-Nr. 51290, als Substrat) 15.000′ Absorptions-U wie vorstehend beschrieben entsprechen ∼1 Bergmeyer-U. 1 Bergmeyer-U ist die Menge an Enzym, die bei einem pH-Wert von 2,0 und 37 °C 1 μmol Acetyl-L-phenyl-3,5-diiod-L-tyrosin hydrolysiert.
