General description
Dithiothreitol (DTT) ist ein Disulfid-Reduktionsmittel (SH), das in Proteinen und in der Aminosäure Cystein vorkommt. Es reduziert eine Disulfidbindung, indem es einen Sechsmembranring bildet und einen Thiol-Disulfid-Austausch einleitet. Es ist ein fast 7-mal stärkeres Reduktionsmittel als βME und daher in niedrigeren Konzentrationen wirksam.
Application
DTT wird wie folgt verwendet:
- als einer der Reaktanten bei der Reduktion und Alkylierung von αs1-Casein, dem Hauptallergen von Kuhmilch′.[1]
- als Bestandteil des Mediums für die Demembranierung und Reaktivierung von Spermien.[2]
- zum Erhalt der Stabilität des Enzyms, da Thiol die aktiven Stellen des Biokatalysators wirksam schützt.[3]
- als Reduktionsmittel, um die Spezifität der Reaktion von N-Ethylmaleimid mit Sulfhydrylgruppen zu testen.[4]
- in der Proteomik-Analyse als In-Solution-Proteinverdauung für die Massenspektrometrie.[5]
- als Pufferkomponente für die Proteinquantifizierung, zur Herstellung von Waschpuffer, Lysepuffer, Probenpuffer und Proteinelutionspuffer.[6][4]
Ein hervorragendes Reagens, um SH-Gruppen in reduziertem Zustand zu halten; reduziert Disulfide quantitativ. DTT-Zugabe in den Probenpuffer ist ein effektives Mittel um Disulfidbindungen in Proteinen vor der Durchführung einer SDS-PAGE zu reduzieren. DTT kann auch dazu benutzt werden, um die Disulfid-Brücken des Crosslinkers N,N′-bis(acryloyl)cystamin zu spalten und dadurch die Matrix eines Polyacrylamidgels aufzulösen. DTT riecht weniger stechend und ist weniger giftig als 2-Mercaptoethanol. In der Regel genügt eine sieben Mal kleinere Konzentration an DTT (100 mM) als die, für 2-Mercaptoethanol notwendige (5% v/v, 700 mM).
Biochem/physiol Actions
Dithiothreitol (DTT) wird in großem Umfang bei der chemischen Peptidsynthese und bei biochemischen Präparationen von Thiolproteinen verwendet. Es wird zudem in Studien zur Proteinchemie verwendet, z. B. zur Proteinfaltung und Aktivität von Enzymen. Es wirkt darüber hinaus als Enzymstabilisator, um die DNA-Modifizierung zu stoppen. DTT vermittelt spezifisch die Thiol-Disulfid-Austauschreaktion, um die intra- oder intermolekularen Disulfidbindungen in Biomolekülen vollständig zu reduzieren. Diese Reaktion führt zur Bildung von Thiolen und dem cyclischen Disulfid von DTT.[7]

Stability and storage data for the reconstituted material is unavailable. Generally, a solution of DTT in Hepes buffer, pH 7.75 is stable for one week at 2 to 8 °C if the container is tightly sealed and the solution is protected from atmospheric oxygen by argon or nitrogen. It is recommended to store the solution for one week at 2 to 8 °C as above or to prepare the solution fresh.

If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.

If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. How long can the powder be left out a room temperature?

The long term storage recommendation for this product is refrigerated at 2-8°C, but is shipped at ambient temperature. Refrigerated products that ship at ambient temperature are stable for up to 2 weeks to allow for transit time. Items left at room temperature for short periods will not be adversely effected.
