General description
Rinderserumalbumin ist ein Modellprotein in verschiedenen Studien und besteht aus 583 Aminosäuren. Es ist ein wasserlösliches Protein mit einer Molekülmasse von 66,4 kDa. Sechs α-Helices bilden drei homologe Domänen von BSA. Je nach pH-Wert durchläuft es eine reversible konformative Isomerisierung. Die native Struktur des Proteins wird bei Erhitzung reaktiv und flexibel.
Application
- Rinderserumalbumin wird in einer Lösung zur Isolierung von Herzmitochondrien verwendet.
- Es wird in Puffer für den Zytotoxizitätsassay verwendet.
- Es wird zur Einleitung einer Nephropathie durch Proteinüberlastung bei Ratten eingesetzt.
Biochem/physiol Actions
Bestimmte konformative und Primärsequenz-Epitope von BSA sind vermutete Allergene bei Menschen mit Rindfleisch- und Milchallergien.
Preparation Note
Dieses oft als Cohn-Fraktion V bezeichnete Produkt wird nach einer modifizierten Methode der Cohn-Fraktionierung mit kaltem Ethanol hergestellt.
Serumalbumin kann auch als Fraktion V bezeichnet werden. Diese Bezeichnungskonvention stammt aus der ursprünglichen Cohn-Methode zur Fraktionierung von Serumproteinen mittels kalter Ethanolfällung. Serumalbumin wurde in der fünften Ethanolfraktion bei der Cohn′schen Methode gefunden. Seither wird Serumalbumin von manchen als „Fraktion V“ bezeichnet, gleichgültig wie es gewonnen wurde. Andere verwenden diese Bezeichnung für Serumalbumin, das durch Ethanolfraktionierungsmethoden gewonnen wurde, die stark modifiziert wurden, seit die ursprüngliche Cohn-Methode beschrieben wurde. Sigma-Aldrich produziert und vertreibt Serumalbumine, die unter Anwendung verschiedener Primärmethoden, einschließlich der ursprünglichen Cohn-Fraktionierungsmethode, modifizierter Ethanolfraktionierungsmethoden, Hitzeschockfraktionierung und Chromatographie aufgereinigt wurden. Weitere Aufbereitungsschritte umfassen ggf. Kristallisierung oder Aktivkohlefiltration.

Enzymatic assay of lipase type XIII from Pseudomonas sp. using a coupled enzyme system of glycerol kinase and glycerophosphate oxidase (EC 3.1.1.3)
Objective: To standardize a procedure for the enzymatic assay of β-Glucuronidase.
To measure hyaluronidase activity, a turbidimetric determination assay is used at 600 nm. One unit of hyaluronidase activity will cause a change in absorbance of 0.330 per minute at pH 5.35 at 37 °C.
This procedure applies to all products from heart muscle that have a specification for L-Lactic Dehydrogenase activity.

The data sheet lists several methods for further purification of BSA after ethanol precipitation but does not say which of these were used for A4503. Is A4503 charcoal treated? The data sheet is the same for other BSA powders (e.g., A2153).

Is this product biotin-free?

This product is not tested for the presence of biotin. However, Albumin products prepared by cold ethanol fractionation are expected to be biotin-free.

Is Product No. A4053, Albumin from bovine serum, sterile? If not, how would recommend sterilizing it?

This product is not necessarily sterile. Solutions of albumin cannot be autoclaved, so in order to sterilize it we would recommend that it be dissolved in the appropriate medium (water, saline or buffered saline), followed by sterile filtration through a low protein-binding membrane.
