Biochem/physiol Actions
Dieser Inhibitor wirkt gegen Trypsin und in geringerem Maß auch gegen Chymotrypsin und Plasmin. Er hemmt auch Proteasen mit ähnlichen Mechanismen wie Trypsin, Plasmakallikrein und Gerinnungsfaktor X. Der Trypsininhibitor wirkt nicht gegen Metallproteasen, gewebebasiertes Kallikrein, Säureproteasen oder Thioproteasen. Dieser Inhibitor wirkt, indem er einen 1:1-stöchiometrischen Komplex mit der proteaseaktiven Stelle bildet und anschließend eine einzelne Arginin-Isoleucin-Bindung an dem Inhibitor spaltet. Die Hemmung ist sowohl reversibel als auch pH-Wert-abhängig.
Preparation Note
Der Trypsininhibitor ist zu 10 mg/ml in Wasser und Phosphatpuffern löslich. Er ist in Balanced Salt Solutions zu 1 mg/ml und in serumfreien Medien löslich. Konzentrierte Lösungen von mehr als 10 mg/ml können trüb und gelblich bis bernsteinfarben sein. Nach der Trypsinisierung von Zellen in 1 ml Trypsininhibitorlösung zu 1 mg/ml für jeden ml Trypsinlösung für die Dissoziation erneut suspendieren. Die Zellsuspension sollte anschließend bei 1.000 U/min zentrifugiert werden, um ein Zellpellet zu bilden.
Lösungen können eine Aktivität behalten, wenn sie kurzzeitig bei 2–8 °C gelagert werden. Lösungen sind in gefrorenen Aliquoten bei -20 °C stabil.

Analysis Note
Ein mg Trypsininhibitor hemmt die Aktivität von mindestens 1,0 mg Trypsin, ca. 10.000 BAEE-Einheiten/mg Trypsin.

Der Sojabohnen-Trypsininhibitor ist ein monomeres Protein, das 181 Aminosäure-Rückstände in einer einzelnen Polypeptidkette enthält, die durch zwei Disulfidbrücken vernetzt ist.
Eine Trypsin-Einheit erzeugt ein ΔA253 von 0,001 pro Minute mit BAEE als Substrat bei einem pH-Wert von 7,6 und einer Temperatur von 25 °C; Reaktionsvolumen 3,2 mL, 1 cm Lichtweg.
Eine Trypsineinheit = A253 von 0,001 je Minute mit N-Alpha-benzoyl-L-argininethylester (BAEE) als Substrat bei einem pH-Wert von 7,6 und 25 °C.

What is the molecular weight and amino acid sequence of trypsin inhibitor from Glycine max (soybean)?

Soybean trypsin inhibitor is a mixture of the three types. We have not characterized our trypsin inhibitor products in terms of how much of each of the three types may be present. All three have a molecular weight of between 20,000 and 20,200 daltons, so one might use 20,100 daltons as an average.The three types vary in the amino acids at a few positions. The detailed summary of the three types (or variants) was published in the following linked reference: J. Biochem. (Japan), 98, 435-448 (1985).Note that the authors use Tia, Tib and Tic to indicate the three variants.
