General description
TMB-Substrat ist ein chromogenes Substrat für Meerrettichperoxidase - HRP.
Application
TMB-Substrat ist für ELISA-Verfahren vorgesehen, die Meerrettichperoxidase-Konjugate einsetzen. TMB-Substrat entwickelt ein lösliches blaues Reaktionsprodukt, das bei 370 oder 655 nm abgelesen werden kann. Die Reaktion wird mit Säure gestoppt, wobei ein gelbes Reaktionsprodukt erzeugt wird, das eine genaue Intensitätsmessung bei 450 nm ermöglicht.

- Unser TMB-Substrat wird als gebrauchsfertiges Peroxidase-Substrat mit TMB in einem schwach sauren Puffer geliefert. Vor der Reaktion mit Peroxidase sollte das Substrat eine farblose bis leicht blau-grüne Lösung sein. Das Substratsystem erzeugt ein blaues Reaktionsprodukt mit Peroxidase.
- Wenn der Assay-Endpunkt durch Zugabe von Säure erreicht ist, messen Sie die TMB-Substratintensität bei 450 nm.
- Unser TMB-Substrat wird für ELISA-Verfahren (Microwell) empfohlen und wurde in verschiedenen Assays als Substrat für Meerrettichperoxidase-Antikörperkonjugat zum Nachweis von Hendravirus-Glycoprotein, Tetracosactid, sIgA und neutrophilen extrazellulären Fallen (NETs) verwendet.
- Das 3,3′,5,5′-3,3,5,5-Tetramethylbenzidin (TMB) Flüssigsubstratsystem wird nicht für Membrananwendungen empfohlen. Da TMB-Substrat ein lösliches Reaktionsprodukt erzeugt, wird es nicht für histochemische oder Blotting-Anwendungen empfohlen.

There is no set time at which the reaction must be stopped. Depending on the peroxidase activity measured, the color may form very quickly or slowly. Kindly adhere to the instructions provided in our datasheet as closely as possible.
