Beschreibung

General description

Guanidinthiocyanat ist ein chaotropes Mittel, das als starkes Proteindenaturierungsmittel dient. Es wird verbreitet in RNA-Isolierungsprotokollen eingesetzt. Guanidinthiocyanat kann RNase und DNase irreversibel inaktivieren und Zellmembranen aufbrechen. Es löst Proteine, indem es ihre Sekundärstrukturen stört. Guanidinthiocyanat trennt außerdem Nukleoproteine von Nukleinsäuren und unterstützt den Aufschluss von Proteinen durch Proteasen. Es steigert die Aktivität von Proteinase K und macht sie dadurch zur bevorzugten Proteinase.

Application

Guanidinthiocyanat-Lösung wurde zum Extrahieren von Folgendem verwendet:

• Plasma-RNA aus mit humanem Immundefizienz-Virus (HIV) infizierten und Kontrollpatienten
• Gesamt-RNA aus gesammeltem Gewebe (Triebe, Blüten, Früchte und Samen)
• in der Reverse-Transkriptase-Polymerase-Kettenreaktion (RT-PCR), RNA aus der Teratokarzinom-Zelllinie (Tera-1)

Disclaimer
