Beschreibung

General description

Ein aus Streptomyces azureus isoliertes, thiazolhaltiges Polypeptid-Antibiotikum. Hemmt die bakterielle Proteinsynthese und ribosomale GTPase-Aktivität durch die nicht-kovalente, aber praktisch irreversible Bindung an die 23S- rRNA im Zentrum der GTPase der 50S-Untereinheit. Durch die direkte Bindung des Thiostreptons wird die Bindung am Ribosom des Elongationsfaktors G (EF-G) verhindert, der auch spezifisch an die 23S-rRNA bindet. Thiostrepton hat sich bei der Aufklärung der molekularen Mechanismen der Translation, insbesondere der Translokation des Ribosoms, als nützlich erwiesen. Es wird auch zur Selektion von Vektoren in Bakterien von Streptomyces sp. verwendet, die das tsr-Gen (S. azureus) enthalten und die eine 23S-rRNA-Methylase kodieren, deren Wirkung die Bindung von Thiostrepton an Ribosomen verhindert.

Ein thiazolhaltiges Peptidantibiotikum, das die Proteinsynthese hemmt, indem es die Bindung von GTP an die 50S-Untereinheit des Ribosoms verhindert. Hemmt die Funktion des Elongationsfaktors G (EF-G) und die Dissoziation von EF-G vom Ribosom. Das thiostreptonresistente Gen Dibutylphthalat (DBP) wird vornehmlich als selektiver Marker für rekombinante DNA-/Plasmidtechnologien verwendet.
