Beschreibung

General description

Elastase ist eine einzelne Polypeptidkette mit 240 Aminosäureresten und enthält vier Disulfidbrücken. Die Molekularmasse beträgt ca. 25,9 kDa. Das Enzym wird als inaktives Zymogen Proelastase synthetisiert, das dann von Trypsin durch beschränkte Proteolyse am N-Terminus in die aktive Form umgewandelt wird.

Application

Elastase vom Pankreas des Schweins wird für folgende Anwendungen eingesetzt:

Biochem/physiol Actions

Elastase hydrolysiert Elastin, das spezifische Protein elastischer Fasern, und verdaut Hämoglobin, Casein und Fibrin.

Enzym für den Abbau von Elastin, einem Matrixbestandteil von Geweben, der an Dehnungsprozessen beteiligt ist. Es handelt sich um eine Serinprotease mit breiter Spezifität. Das Enzym spaltet Protein auf der Carboxylseite von kleinen hydrophoben Aminosäuren wie Ile, Gly, Ala, Ser, Val, und Leu auf. Ebenso hydrolysiert es Amide und Ester wie etwa N-Benzoyl-L-Alanin-Methylester.
