General description

Dulbecco′s Modified Eagle′s Medium (DMEM), das regelmäßig als Zellkulturmedium verwendet wird, ist eine Quelle für energiereiche Medien, die Glucose, Vitamine und Aminosäuren umfasst.[1] Eine 1:1-Mischung von Dulbecco′s Modified Eagle′s Medium (DME) und Hams F-12-Nährstoffmischung wird verbreitet in der Zellkulturforschung eingesetzt. Um den durch das Weglassen des Serums ausgelösten Verlust der Pufferkapazität auszugleichen, kann den Formulierungen eine Endkonzentration von 15 mmol/l 4-(2-Hydroxyethyl)-1-piperazinethansulfonsäure(HEPES)-Puffer zugegeben werden.

Application

Dulbecco′s Modified Eagle′s Medium/Nährstoffmischung F-12 nach Ham:
  • wurde als Komponente im Kulturmedium zum Kultivieren von primären kulturaktivierten humanen Lebersternzellen eingesetzt[1]
  • wurde zum Kultivieren von Knorpelbiopsien eingesetzt[2]
  • kann zum Propagieren von Cowdria ruminantium eingesetzt werden[3]

  2. Is the glutamine that has already been added stable glutamine? How long is the shelf life?

This product does not contain Alanyl-glutamine, which is considered a more stable form It has been supplemented with L-glutamine and will remain stable through out the shelf life of the product with proper storage and handling. Typical lots of D8437 are assigned an expiration date of 1 year from the date of release. Note that under culture conditions of 37 deg C the L-glutamine will gradually degrade.     1. This is a 1:1 mixture of the DME and F-12. The optimal carbon dioxide required for DME is 10% and for F-12 is 5%. Since this medium is a mixture, the optimal carbon dioxide concentration is typically 5% to 8%.

    1. When DME/F12 is used under serum free conditions, supplementation is necessary. DME/F12 can be used for growing the epithelial cell line TM4 (mouse testes) when supplemented with insulin, transferrin, EGF, FSH, somatomedin C, ovine growth hormone, and retinoic acid. Primary cultures of Sertoli cells can also be grown in the above supplemented media. Rat glial C6 cells have been grown in DME/F12 supplemented with transferrin, insulin, LH, and rat submaxillary gland extract.
