Beschreibung

General description

Aprotinin ist ein Protein mit einer Länge von 58 Aminosäuren und einem berechneten Molekulargewicht von 6512 Da, das als Serinprotease-Inhibitor agiert. Aprotinin, auch als boviner pankreatischer Trypsin-Inhibitor (BPTI) bekannt, hemmt Serinproteasen wie Chymotrypsin, Kallikrein, Plasmin und Trypsin.

Application

Aprotinin wird im Wesentlichen als Trypsin-Inhibitor eingesetzt.

Biochem/physiol Actions

Aprotinin ist ein kompetitiver Inhibitor der Serinprotease, der stabile Komplexe mit Enzymen bildet und aktive Enzymstellen blockiert. Diese Bindungen sind reversibel und die meisten Aprotinin-Protease-Komplexe lösen sich bei extremen pH-Werten von >10 oder <3 auf. Strukturell ist Aprotinin ein monomeres globuläres Protein; ein Derivat aus der Rinderlunge, das aus 58 Aminosäuren besteht, die in einer einzelnen Polypeptidkette mit drei vernetzenden Disulfidbrücken angeordnet sind.

Preparation Note

Bei diesem Produkt handelt es sich um eine aseptisch filtrierte Lösung in 0,9 % NaCl und 0,9 % Benzylalkohol. Es hat eine Aktivität von 3–7 TIU/mg in Protein und and 3–21 TIU/ml in Lösung.

Other Notes

Eine Trypsin-Inhibitor-Einheit (TIU) senkt die Aktivität von zwei Trypsin-Einheiten um 50 %, wobei eine Trypsin-Einheit bei pH 7,8 und 25 °C 1,0 μmol N-α-Benzoyl-DL-arginin-p-nitroanilid pro Minute hydrolisiert. Eine weitere häufig verwendete Einheit ist KIU, wobei 1 TIU = 1.300 KIU.
