Application
Aprotinin wird im Wesentlichen als Trypsin-Inhibitor eingesetzt.
Aprotinin aus Rinder-Lungengewebe wird bei Insulin- und Glucagon-like Peptide-1-Assays verwendet. Es wird auch bei Western Blots verwendet.[1]
Biochem/physiol Actions
Aprotinin ist ein kompetitiver Inhibitor der Serinprotease, der stabile Komplexe mit Enzymen bildet und aktive Enzymstellen blockiert. Diese Bindungen sind reversibel und die meisten Aprotinin-Protease-Komplexe lösen sich bei extremen pH-Werten von >10 oder <3 auf. Strukturell ist Aprotinin ein monomeres globuläres Protein; ein Derivat aus der Rinderlunge, das aus 58 Aminosäuren besteht, die in einer einzelnen Polypeptidkette mit drei vernetzenden Disulfidbrücken angeordnet sind.
Aprotinin ist ein natürliches Polypeptid aus 58 Aminosäuren. Es hat hämostatische/Antiplasmin-Eigenschaften und trägt bei chirurgischen Eingriffen zur Reduzierung des Blutverlustes bei. So wird der Bedarf an Bluttransfusionen verhindert.
Preparation Note
Dieses Produkt ist ein lyophilisiertes Pulver, das zellkulturgetestet wurde. Es hat eine Aktivität von 3–7 TIU/mg.
Other Notes
Eine Trypsin-Inhibitor-Einheit (TIU) senkt die Aktivität von zwei Trypsin-Einheiten um 50 %, wobei eine Trypsin-Einheit bei pH 7,8 und 25 °C 1,0 μmol N-α-Benzoyl-DL-arginin-p-nitroanilid pro Minute hydrolisiert. Eine weitere häufig verwendete Einheit ist KIU, wobei 1 TIU = 1.300 KIU.
Wenn Benzoyl-L-Argininethylester (BAEE) als Substrat verwendet wird, entspricht eine BAPNA-Einheit etwa 45 BAEE μmolaren Einheiten bei pH 8,0 und 25 °C oder etwa 9.000 BAEE A253 Einheiten bei pH 7,6 und 25 °C.
