Beschreibung

General description

Am häufigsten verwendeter Phorbolester. Hochwirksamer Tumorpromoter auf der Maushaut. Aktiviert Proteinkinase C in vivo und in vitro, auch bei Konzentrationen im nM-Bereich. Fördert die Expression von induzierbaren NOS in gezüchteten Hepatozyten. Aktiviert die Ca²⁺-ATPase und potenziert die Forskolin-induzierte cAMP-Bildung. Hemmt die vom Fas-Antigen induzierte Apoptose, aber induziert die Apoptose in promyelozytischen HL-60-Leukämiezellen. Seine Bindung ist reversibel. Auch als 10-mM-Lösung in DMSO verfügbar (Katalog-Nr. 500582).

Preparation Note

Nach der Rekonstituierung in Aliquote aufteilen und einfrieren (-20 °C). Wasserfreie DMSO-Stammlösungen sind bei -20 °C bis zu 6 Monate haltbar. Bei häufiger Verwendung kann sich in den DMSO-Stammlösungen Wasser ansammeln. Wenn in der Stammlösung Wasser vorhanden ist, beschleunigt sich der Abbau des produktes, was in 2 bis 3 Monaten eine signifikante Aktivitätsminderung zur Folge haben kann.

Unmittelbar vor Gebrauch mit wässrigen Pufferlösungen verdünnen.
