Beschreibung

General description

Ein strukturelles Torin1-Analogon (Katalog-Nr. 475991), das als zellpermeabler, potenter, ATP-kompetitiver Inhibitor gegen DNA-PK und mTOR wirkt (IC₅₀ = 0,5 bzw. 2,81 nM). Im Vergleich zu Torin1 zeigt Torin2 eine stärkere hemmende Wirkung gegen PI 4-Kβ (IC₅₀ = 18,3 nM) und PI 3-K (IC₅₀ = 4,68, 5,67, 8,58, 17,5, 24,5 und 28,1 nM, jeweils gegen P110α/P85α, P110γ, hVPS34, P110δ/P85α, C2β, C2α), während es bei der Hemmung der S6K1(Thr389)-Phosphorylierung in MEF-Kulturen in vitro wirksamer ist (IC₅₀ = 0,25 nM vs. 2 nM, jeweils für Torin2 und Torin1), und es zeigt in vivo eine viel bessere orale Bioverfügbarkeit bei Mäusen (C_max (ng/ml)/T_1/2 (h) = 3968/0,72 bzw. 223/0,79, jeweils für Torin2 und Torin1; 10/mg/kg p. o.).
