Beschreibung

General description

Ein zellpermeables Pyridinon-Chinolin-Derivat, das als hochpotenter, ATP-kompetitiver Inhibitor gegen mTOR und DNA-PK (IC₅₀ = 4,32 bzw. 6,34 nM in zellfreien Kinase-Assays) wirkt und PI 3-K der Klasse I (IC₅₀ = 171, 250 und 564 nM gegen P110γ, P110α/P85α bzw. P110δ/P85α), der Klasse II (IC₅₀ = 176 und 564 nM gegen C2α bzw. C2β) und der Klasse III (IC₅₀ = 533 nM gegen hVPS34) erst bei viel höheren Konzentrationen hemmt. Hemmt in vitro wirksam die mTORC1-vermittelte S6K1-Thr389-Phosphorylierung in MEF-Kulturen (IC₅₀ = 2 nM) sowie in vivo die mTORC2-vermittelte Akt-Ser473- und die mTORC1-abhängige S6-Ser235/236-Phosphorylierung in der Lunge und der Leber von Mäusen (bis zu 6 Stunden nach einer einmaligen intraperitonealen Dosis von 20 mg/kg). Berichten zufolge unterdrückt Torin1 trotz seiner geringen In-vivo-Stabilität (T_1/2 = 4,52 Stunden bei Mäusen; 10 mg/kg intraperitoneal) die Ausbreitung von U87MG-Tumoren bei Mäusen vollständig, wenn es in einer hohen Dosierung von 200 mg/kg täglich intraperitoneal verabreicht wird. Zudem wird berichtet, dass es wirksam zelluläre mTORC1-Funktionen hemmt, die als Rapamycin-resistent gelten (Katalog-Nr. 553210, 553211 und 553212M).
