As indicated above, this product is soluble in distilled water to 10 mg/mL. Following reconstitution, elastase may be stored at 4°C at pH 6.0 for long term use. If incubated at room temperature at or near its optimum pH, elastase rapidly autolyzes to a mixture of peptides. Stock solutions are stable for up to 2 months at 4°C, pH 6.0.

General description
Native Elastase vom Pankreas des Schweins. Diese Serinprotease katalysiert die Hydrolyse von Proteinen und Peptiden – darunter Albumin, Casein, denaturiertes Kollagen, Elastin, Fibrin und Hämoglobin – und von einer Reihe synthetischer Substrate, die Asparaginsäure, Phenylalanin oder Tyrosin enthalten, insbesondere an Bindungen, die neutralen Aminosäureresten benachbart sind. Gehemmt von DFP, Elastinal und α2-Makroglobulin.

Spaltet die Peptidbindungen vorzugsweise am Carbonylende von Aminosäureresten mit kleinen hydrophoben Seitenketten, etwa Glycin, Valin, Leukin, Isoleukin und ganz besonders Alanin. Gehemmt von DFP, Elastinal und α2-Makroglobulin. Der optimale pH-Wert ist 7,8–8,5; pI = 9,5. Hinweis: Elastase neigt zum Anhaften an Glas. Daher wird die Verwendung silikonisierter Glaswaren empfohlen.

Lyophilisiert aus 50 mM Trehalose, 1 mM Essigsäure.
Preparation Note
Nach der Rekonstitution kann Elastase für die langfristige Verwendung bei 4 °C und pH 6,0 gelagert werden. Wenn Elastase bei Raumtemperatur und optimalem (oder fast optimalem) pH-Wert inkubiert wird, erfolgt rasch eine Autolyse in eine Peptidmischung. Stammlösungen sind bei 4 °C und pH 6,0 bis zu 2 Monate haltbar.
