Das Molverhältnis von Protein A:Biotin wird auf chargenspezifischer Basis bestimmt. Die Bindungsstärke ist wie folgt: stark reaktiv: Meerschweinchen, humanes IgG1, IgG2 und IgG4 sowie Kaninchen; schwach reaktiv: Rind, humanes IgA, IgM und IgE sowie Maus; nichtreaktiv: Ziege, Pferd, humanes IgD und IgG3, Ratte und Schaf. o-Phenylendiamin, 1,0 mg/ml in Citratpuffer, pH-Wert 4,5, 0,1 % Harnstoffperoxid bei 25 °°C wurde als Substrat zum Prüfen der Funktionalität von Biotin-Protein A und Avidinperoxidase eingesetzt. Mit den Assay-Bedingungen verbundene Variablen geben die korrekte Arbeitsverdünnung vor.
