General description

Protein A is a 42 kD, highly stable cell surface receptor isolated from Staphylococcus aureus which binds selectively and with high affinity to the Fc portion of IgG from various species[1]. It can also be immobilized on solid support like agarose or acrylic beads and is used for purification of monoclonal or polyclonal immunoglobulins. Protein A reacts specifically to IgG of monkey, rabbit, pig, guinea pig, dog, and cat[2].

Application

Protein A may be used for immunoprecipitation assays[3] [4] . It can be conjugated to fluorescent dyes, enzyme markers, biotin or colloidal gold and can be used in immunohistochemistry, western blotting and ELISA.

Protein A kann mit verschiedenen Reporter-Molekülen konjugiert werden, darunter mit fluoreszierenden Farbstoffen (FITC), Enzymmarkern (Peroxidase, β-Galactosidase, alkalischer Phosphatase), Biotin und kolloidalem Gold, ohne dass sich dies auf die Antikörper-Bindungsstelle auf dem Molekül auswirkt. Diese Konjugate werden zum Nachweis von Immunglobulinen in verschiedenen immunchemischen Assays verwendet, u. a. im Western Blot, in der Immunhistochemie und in ELISA-Anwendungen. Darüber hinaus kann Protein A zur Aufreinigung von polyklonalen oder monoklonalen Immunglobulinen auf einem festen Träger wie Agarose- oder Acryl-Beads immobilisiert werden. Es wird auch routinemäßig für Immunpräzipitations-Assays verwendet.

Protein A ist ein hochstabiler Zelloberflächenrezeptor, der von mehreren Stämmen von Staphylococcus aureus erzeugt wird. Er besteht aus einer einzelnen Polypeptidkette mit einem Molekulargewicht von 42 kDa, die vier repetitive Domänen enthält, die reich an Asparagin- und Glutaminsäure, aber frei von Cystein sind. Er enthält wenig oder keine Kohlenhydrate, nur 4 Tyrosinreste und keine Tryptophane. Protein A kann am Fc-Abschnitt von Immunglobulinen, insbesondere IgGs, einer großen Zahl von Spezies binden. Es wurde nachgewiesen, dass ein einzelnes Protein-A-Molekül mindestens 2 Moleküle von IgG gleichzeitig bindet. Die IgG-bindende Domäne von Protein A besteht aus drei antiparallelen α-Helices, deren dritte disruptiert wird, wenn das Protein mit dem Fc-Abschnitt der Immunglobuline komplexiert wird. Protein A bindet den Fc-Abschnitt von humanen IgG-Unterklassen, IgM, IgA und IgE; und von murinem IgG1 (schwach), IgG2a und IgG2b. Weiterhin bindet Protein A IgGs von anderen Spezies, darunter Affe, Kaninchen, Schwein, Meerschweinchen, Hund und Katze.
Protein A ist auch an mehreren verschiedenen biologischen Schutzfunktionen beteiligt, darunter an Antitumor-, toxischen und karzinogenen Aktivitäten. Es fungiert nicht nur als Immunmodulator, sondern besitzt auch antimykotische und antiparasitäre Eigenschaften.

Physical form

~10% (wet weight/volume) of essentially non-viable S. aureus (Cowan strain) cells in 0.04 M potassium phosphate buffer, pH 7.2, 0.15 M sodium chloride containing 0.2% sodium azide.
