General description

Das Bactident^™ Coagulase-Reagens ermöglicht den zuverlässigen Nachweis des Enzyms Koagulase von Staphylococcus aureus. Koagulase ist ein Enzym, das Plasma koagulieren kann. Staphylococcus aureus bildet zwei Typen von Koagulasen; (i) die freie Koagulase ist ein extrazelluläres Enzym und (ii) die gebundene Koagulase ist auf der Oberfläche der Zellwand lokalisiert. Mit dem Röhrchentest können beide Enzyme nachgewiesen werden, wohingegen der Objektträgertest nur die gebundene Koagulase erkennt.

Application

Eine Übernacht-Bouillonkultur von Staphylococcus aureus wird mit rehydriertem EDTA-Kaninchenplasma inkubiert, das in Bactident^™ Coagulase enthalten ist. Der Koagulasetest ist positiv, wenn mehr als drei Viertel des Röhrcheninhalts ein zusammenhängendes Gerinnsel bilden.
