General description

Isoelektrischer Punkt (pl): 4,7 - 5

Biochem/physiol Actions

Wandelt Tyrosin in L-DOPA oder Tyramin in Dopamin um

Questions

1–10 of 14 Questions
  1. Is it possible to use an equivalent buffer, such as phosphate buffer (Na/K), instead of the recommended potassium phosphate buffer to reconstitute the enzyme?

    1 answer
    1. Tyrosinase is soluble (2 mg/mL) in 50 mM potassium phosphate buffer, pH 6.5, yielding a clear brown solution.
The use of other buffers, such as phosphate buffer has not been evaluated. For optimal results and to minimize the risk of issues, we recommend closely following the provided instructions.

  2. Is it possible to reconstitute T3824-25KU with a solution other than potassium phosphate buffer?

    1 answer
    1. The enzymatic assays are performed using potassium phosphate buffer as a solvent. Unfortunately, no other solvent was tested in-house. An online search indicates that water may be used to initially reconstitute the enzyme and then further dilute to the solution suitable for the application.

  3. Does the solid contain any copper specie?

    1 answer
    1. Yes, all native forms of Tyrosinase from mushroom contain Copper. Please see the link below to review additional information regarding the structure and sequence information of Tyrosinase from mushroom.

  5. When measuring the activity of Tyrosinase, should we use the highest change observed every minute between Tyrosinase and the mixture (with L-tyrosine) as the activity, or the average change over the measured time?

    1 answer
    1. Refer to the protocol of the enzymatic assay of Tyrosinase as described in the reference: Duckworth, H. W. and Coleman, J. E. (1970) Journal of Biological Chemistry, 245, 1613-1625. The protocol can be accessed at https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/776/246/tyrosinase.pdf

  6. Is this tyrosinase is derived from Agaricus bisporus or another mushroom species?

    1 answer
    1. Yes, this product is derived from Agaricus bisporus.

  7. Can you please tell me 25000 units tyrosinase in milligrams?

    1 answer
    1. This product is sold by activity. The minimum activity specification for this material is 1000 units per milligram. The weight of this product can be determined using the activity value reported on the lot specific Certificate of Analysis(COA). For example, the sample COA has a activity value of 6540 units/mg. The amount of powder in the 25,000 unit package will be 3.82mg.

Please see the link below or navigate to the ‘DOCUMENTATION’ section of the Product Detail Page to access a sample or lot specific Certificate:
