Glucose-Oxidase aus Aspergillus niger ist ein Dimer, das aus 2 gleichen Untereinheiten mit einem Molekulargewicht von je 80 kDa besteht. Jede Untereinheit enthält einen Flavinadenindinukleotid-Anteil und ein Eisen. Das Enzym ist ein Glycoprotein, das ~ 16 % Neutralzucker und 2 % Aminozucker enthält. Das Enzym enthält auch 3 Cysteinreste und 8 potenzielle Stellen für die N-verknüpfte Glykosylierung.
Glucose-Oxidase kann D-Aldohexosen, Monodesoxy-D-Glucosen und Methyl-D-Glucosen mit unterschiedlichen Geschwindigkeiten oxidieren.
Der optimale pH-Wert für die Glucose-Oxidase ist 5,5, doch der Aktivitätsbereich erstreckt sich von pH 4 bis pH 7. Glucose-Oxidase ist spezifisch für β-D-Glucose mit einem KM von 33–110 mM.
Glucose-Oxidase erfordert keine Aktivatoren, wird aber von Ag+, Hg2+, Cu2+, Phenylquecksilberacetat und p-Chlorquecksilberbenzoat gehemmt. Es erfolgt keine Hemmung durch die nichtmetallischen SH-Reagenzien: N-Ethylmaleimid, Iodacetat und Iodacetamid.
Glucose-Oxidase eignet sich für die enzymatische Bestimmung von D-Glucose in einer Lösung. Da Glucose-Oxidase β-D-Glucose zu D-Gluconolacton und Wasserstoffperoxid oxidiert, wird für die Glucose-Bestimmung oft Meerrettichperoxidase als Kopplungsenzym verwendet. Obwohl Glucose-Oxidase für β-D-Glucose spezifisch ist, können D-Glucose-Lösungen quantifiziert werden, da die α-D-Glucose eine Mutarotation zu β-D-Glucose erfährt, während die β-D-Glucose durch die enzymatische Reaktion konsumiert wird.

According to the size of the container that you describe for each product, how can I estimate its size in grams?

This product is sold by "units of activity". The activity specification for this item is 100,000-250,000 units/g solid. The exact result will be reported on the lot-specific Certificate of Analysis.
For example, lot SLCR6049 has a specific activity of 151,821 units/gram. Dividing the 10,000 unit pack size by the lot-specific activity will reveal the mass weight within the vial - the 10KU package size for this lot contains 65.8 µg.

How should solutions of Glucose oxidase, Product G7141, be stored?

No solution stability studies have been performed by Sigma. However, we would expect solutions prepared in buffer in the pH range of 5-7 to be stable at 2-8°C for at least one week. For longer term storage, solutions can be stored as frozen aliquots at -20°C (only one freeze/thaw cycle) for at least 6 months.
