General description
Chemische Struktur: Indol-Derivat
Staurosporin ist ein Alkaloid und wird aus Streptomyces sp. gewonnen. Es verfügt über ein breites Spektrum an Eigenschaften, darunter die Hemmung der Proteinkinase C und der Tumorzellproliferation sowie Blockierung der Zellzyklusprogression in verschiedenen Zellen.[1] Staurosporin ist ein bekannter Induktor der Apoptose in verschiedenen Zelllinien. Die Mechanismen der durch Staurosporin induzierten Apoptose variieren bei verschiedenen Zelllinien, darunter mitochondriale apoptotische Signalwege, Bcl-2-Überexpression und caspaseunabhängige Mechanismen.[2] Es macht die Multiresistenz (Multi-Drug-Resistence, MDR) teilweise rückgängig, indem es Zellen des MDR-Phänotyps für zytotoxische Wirkstoffe sensibilisiert. Staurosporin hemmt die Phosphorylierung des p-Glykoproteins (PGP). Die funktionelle Signifikanz der PGP-Phosphorylierung ist jedoch nicht hinreichend geklärt.
Application
Staurosporin wird als Apoptose-Induktor bei Apoptose-Assays an humanen Zelllinien des Kolorektalkarzinoms[3] und zur Beurteilung der Caspase-3- und Caspase-7-Aktivität in Endothelzelllinien verwendet.[4] Es wird zudem als Positivkontrolle im Apoptose-Assay an humanen Endothelzellen der Nabelvene eingesetzt.[5]

Starker Inhibitor der Calcium/Phospholipid-abhängigen Proteinkinase. Hemmt die Hochregulierung der VEGF-Expression in Tumorzellen.
