General description

C1q bildet zusammen mit C1r und C1s im Verhältnis 1:2:2 den C1-Komplex, der die erste Komponente des klassischen Komplementwegs darstellt. C1q besteht aus 18 Polypeptidketten (sechs A, sechs B, sechs C) (Molekulargewicht: 460 kDa). Alle enthalten einen Collagen-ähnlichen 81-aa-Bereich, der aus sich wiederholenden Sequenzen (Gly-Xaa-Yaa) nahe dem N-Terminus besteht. Drei Ketten (A1B1C1) bilden eine Dreifachhelix mit dem C-Terminus als globulären Kopf, der strukturell und funktionell unterschiedliche Domänen enthalten kann.

Application

Die Komplementkomponente C1q ist ein wichtiger Regulatorfaktor für die Plättchenaktivierung. Mit diesem Thema befasst sich die Forschung seit einiger Zeit, da Thrombozyten-Leukozyten-Aggregate bei inflammatorischen Erkrankungen, wie der koronaren Herzerkrankung, eine wichtige Rolle spielen. Insbesondere wurde nachgewiesen, dass C1q die Collagen-induzierte Aggregation hemmt und die Produktion von reaktiven Sauerstoffspezies (ROS) verbessert.

Biochem/physiol Actions

Eine C1q-Defizienz bedingt durch einen seltenen homozygoten Gendefekt oder transkriptionelle/translationale Defekte ist höchstwahrscheinlich eine Ursache des systemischen Lupus erythematodes (SLE). Die drei C1q-Untereinheiten werden durch einen neuartigen Mechanismus der transkriptionellen Synchronisation exprimiert.

    1. The complement is supplied in solution at a concentration of approximately 1 mg/mL, with the lot-specific certificate of analysis providing the exact concentration. Solubility is tested by diluting 0.1 mL of the complement stock solution with 0.2 mL of H2O, resulting in a final volume of 0.3 mL. This dilution ratio yields a complement concentration of approximately 0.33 mg/mL.

    1. This product corresponds to purified 'native' human C1q. Its sequence is not determined in-house. Please refer to published sequences, e.g. in GenBank for UniProt accession nos. P02745, P02746, & P02747.

  3. what is the size of the protein in WB ? when I test it's size it was approximately 24 KD ( the same size as c1q-c )

    1. C1q is composed of 18 polypeptide chains (six A, six B, six C). When prepared in denaturing sample buffer, and run on SDS PAGE, the chains will likely dissociate and run at their individual molecular weights. The Uniprot entries for C1qA, C1qB, and C1qC list masses of 26,017, 26,722, and 25,774, respectively.
