General description

Alkoholoxidase (AOX) ist ein Homooktamer mit einer Molekülmasse von 600 kDa und acht Flavin-Adenin-Dinukleotid(FAD)-Cofaktoren. Es gehört zur Glucose-Methanol-Cholin(GMC)-Oxidoreduktasefamilie.

Application

Eine Alkoholoxidase-Lösung aus Pichia pastoris wird im Baumwoll-Pektinmethylesterase(PME)-Enzym-Assay verwendet. Darüber hinaus wird sie als visueller Biosensor im Teststäbchen-Format zur Immobilisierung auf Polyanilin(PANI)-Film für den Nachweis von Ethanol in fermentierten Getränkeproben verwendet.

Alkoholoxidase kann zur Untersuchung der Proteintranslokation zu Peroxisomen verwendet werden. Produkt A2404 stammt aus Pichia pastoris. Es wird zur Bakterienexpression und immunologischen Verifizierung von Hv-p68-cDNA-Klonen verwendet[1].

Biochem/physiol Actions

Alkoholoxidase katalysiert die Oxidation von kurzkettigen, primären, aliphatischen Alkoholen zu ihren jeweiligen Aldehyden. Es weist die höchste Affinität zu Methanol auf. Alkoholoxidase ist am Stoffwechsel von Methanol beteiligt, was für Hefen von Bedeutung ist, bei denen Methanol die einzige Energiequelle darstellt. Alkoholoxidase ist vornehmlich im Peroxisom anzutreffen, ist ggf. aber auch im Zytoplasma vorhanden.
