General description

Alizarin Red, an anthraquinone derivative, may be used to identify calcium in tissue sections and cultured cells in vitro. The reaction is not strictly specific for calcium, since magnesium, manganese, barium, strontium, and iron may interfere, but these elements usually do not occur in sufficient concentration to interfere with the staining. Calcium forms an Alizarin Red S-calcium complex in a chelation process, and the end product is a bright red stain. Alizarin red is a commonly used stain to identify calcium containing osteocyte in differentiated culture of both human and rodent mesenchymal stem cells.

    1. This product is presented as a 40 mM solution in distilled water, at pH 4.1 ± 0.1, and is ready-to-use. The recommended volume is 1 ml for a 24-well plate and 0.5 ml for a 48-well plate.

    1. Products TMS-008 and 2003999 are similar products. Product TMS-008 is presented as a 40mM solution at pH 4.1 +/- 0.1.

      Product 2003999 is also a 40mM solution with pH range 4.1 +/- 0.1. It is also a kit component of Product ECM815.

    1. Regarding your inquiry about TMS-008-C Alizarin-Red Staining Solution, there is currently no protocol available on sigmaaldrich.com for staining cells with TMS-008. The following information does appear on the emdmillipore.com website:

      Product is presented as a 40mM solution at pH 4.1 +/- 0.1.

      As the molecular weight is 342.26, the percentage of Alizarin Red should be 1.4%. Most of the common histology textbooks generally recommend using a 2% concentration of Alizarin Red. But as in many histology procedures, the exact percentage is generally not critical for acceptable staining. It would appear the developers of the solution thought they preferred a concentration of 1.4%. This is typical of many histological staining procedures. Methods can and do vary from source to source.

      As to a protocol, protocols for most histology textbooks should be acceptable for use. In general, tissue should not be fixed in fixative that contain calcium. Fixatives commonly used as 10% Neutral Buffered Formalin or alcoholic formalin. Once the slides have been prepared, hydrate sections in 50% alcohol. Then rinse briefly with distilled water. Incubate the slides in the Alizarin Red solution anywhere from 30 second to 5 minutes or more. Check for stain development every 30 seconds under a microscope until a red lake forms. The lake should not be too diffuse and on the heavy side. Then remove the solution by shaking off the excess and blot dry. Flood slides with acetone for about 20 seconds. Use a 50:50 mixture of acetone and xylene to differentiate. Clear in xylene. If possible, mount with a synthetic mounting media.

      The solution does not have great stability. The website further states: Store at 2-8ºC for up to 4 months from date of receipt. If Alizarin-red solution forms a precipitate, remove particulates by passage through a 0.22 or 0.45-micron filter.
