General description

Iron, the most abundant transition metal in our bodies, is involved in a number of biologically important processes such as respiration, oxygen transport, and energy production in collaboration with oxygen. In living cells, iron exists mainly as ferrous (Fe2+) and ferric (Fe3+) ionic forms. As a major contributor to oxidative damage of cells, Fe2+ is implicated in serious diseases such as cancers and neurodegenerative disorders, because of its ability to produce harmful reactive oxygen species via contact with oxygen, superoxide, and hydrogen peroxide (H2O2).BioTracker FerroOrange Live Cell Dye is an orange fluorescent probe that specifically detects labile iron (II) ions (Fe2+) only. The intensity of fluorescence does not increase in the presence of iron (III) ions (Fe3+) or bivalent metal ions other than iron. It also does not react to chelated iron in ferritin and other substances. FerroOrange is suitable for live-cell imaging because it is highly cell-permeable and has low cell toxicity.Spectral Properties:Absorbance (max): 542 nmEmission (max): 572 nmResearch Category:Live Cell Dye

Questions

1–5 of 5 Questions
  1. How many tests can be carried out using the 35 nmol package please?

    1 answer
    1. The number of 'tests' is dependent upon the culture vessel and will have to be determined by the end user. Chamber slides or coverslips will require less volume that t-flasks or culture dishes. As per instructions, the prepared 1mM stock solution is diluted to a working concentration of 1 µM prior to applying to cells. Please see the link below to review these instructions and a general protocol provided in the product datasheet:

  2. Can this kit be used for suspension cells?

    1 answer
    1. Yes, this product is suitable for use with suspension cells.

  3. Is it possible to detect SCT210 FerroOrange by flow cytometry?

    1 answer
    1. The website indicates an excitation wavelength of 542 nm and an emission wavelength of 572 nm for the SCT210 FerroOrange. The product is claimed to be suitable for live cell imaging, which typically implies the use of a confocal microscope. However, given that this is a fluorescent dye, it seems reasonable to consider that the staining could be assessed using a flow cytometer. There is currently no available data documenting its use on a flow cytometer. To use it on a flow cytometer, the cells would need to be in a single cell suspension. Additionally, it is expected that the cells could be visualized using a fluorescent microscope.

  4. Is it possible to store SCT210 after reconstitution?

    1 answer
    1. The available datasheet for SCT210 provides guidance in the Reagent Preparation section, stating, "Use neutral buffer or culture medium for dilution of the DMSO solution and use immediately upon dilution. The dye could be oxidized in acidic solutions." However, there are no recommendations or data suggesting that the solutions are stable when stored. It's important to note that the package is shipped in single-use vials.

  5. What cells can be used as a positive control for SCT210 (BioTracker™ FerroOrange Live Cell Dye)?

    1 answer
    1. The HepG2 cell line is recommended as a positive control for SCT210 (BioTracker™ FerroOrange Live Cell Dye).
